Dacarbazine is a synthetic antineoplastic agent classified as an alkylating agent, primarily used in the treatment of malignant melanoma, Hodgkin lymphoma, and soft tissue sarcoma. It is administered intravenously. Dacarbazine acts mainly by methylating and cross-linking DNA after hepatic activation to its active metabolite (MTIC), thereby inhibiting DNA, RNA, and protein synthesis. It may also function as a purine analog and interact with sulfhydryl groups. The drug was first synthesized at Southern Research in Alabama in 1959 and received FDA approval in 1975. Generic versions are widely available[1][3][4][6][7].
