This is a combination chemotherapy regimen consisting of two antineoplastic agents—dacarbazine and mitomycin. Dacarbazine is an alkylating agent that acts as a prodrug; it undergoes metabolic activation in the liver to form active methylating species that inhibit DNA synthesis by causing DNA strand breaks and cross-linking. Mitomycin is an antitumor antibiotic derived from Streptomyces caespitosus; it functions primarily as an alkylating agent that cross-links DNA strands, inhibiting DNA synthesis and leading to cell death. Both drugs are cell cycle phase-nonspecific but have maximal effects at different phases. This combination has been studied for use in advanced colorectal cancer and metastatic gastrointestinal adenocarcinoma but has shown limited efficacy in these settings[2][4]. The primary indications for the individual drugs include malignant melanoma and Hodgkin's disease (dacarbazine) as well as various solid tumors such as gastric, pancreatic, bladder, anal, cervical cancers (mitomycin)[3][5][7].
