Overview

**Dacarbazine + thymosin alpha 1** is a combination therapy comprising dacarbazine, a classic alkylating antineoplastic agent, and thymosin alpha 1 (also known as thymalfasin), an immunomodulatory peptide. Dacarbazine exerts direct cytotoxic effects on tumor cells by alkylating DNA, leading to apoptosis and inhibition of cell division. Thymosin alpha 1, a 28-amino acid peptide, enhances host immune responses primarily by promoting T-cell differentiation and activation, stimulating cytokine production (e.g., IFN-γ, IL-2), boosting natural killer (NK) cell activity, and upregulating toll-like receptors (e.g., TLR2 and TLR9) in dendritic cells. The combination has been most extensively studied in the context of metastatic melanoma, where it aims to both directly kill tumor cells (via dacarbazine) and enhance anti-tumor immunity (via thymosin alpha 1). Clinical trials indicate that this combination may improve response rates and survival without significant increases in toxicity compared to standard therapy[1][4][7].
