Overview

Dacetuzumab is a humanized IgG1 monoclonal antibody that targets the CD40 antigen, which is highly expressed on various hematological malignancies including multiple myeloma, non-Hodgkin lymphoma, chronic lymphocytic leukemia, and diffuse large B-cell lymphoma. It binds to the CD40 receptor and acts as a partial agonist without blocking CD40/CD40L interactions. Dacetuzumab induces tumor cell death through several mechanisms: direct induction of apoptosis via signal transduction, antibody-dependent cellular cytotoxicity (ADCC), and phagocytosis (ADCP). It also enhances immune effector functions against tumor cells. The drug was developed by Seattle Genetics (now Seagen) and licensed to Genentech for further development. Clinical trials were conducted in various hematologic cancers but development has been discontinued for these indications[1][2][3][4][6].
