Daclizumab + exenatide is an experimental drug combination consisting of daclizumab, a humanized monoclonal antibody targeting the interleukin-2 receptor (CD25), and exenatide, a glucagon-like peptide 1 (GLP-1) receptor agonist. This combination was investigated in patients with long-standing type 1 diabetes to assess whether it could improve residual beta-cell function by both suppressing autoimmune activity (daclizumab) and promoting beta-cell recovery or regeneration (exenatide). Clinical studies found that while exenatide reduced body weight and insulin requirements, the combination did not result in improved endogenous insulin secretion or beta-cell function. Daclizumab acts as an immunosuppressant by blocking IL-2-mediated T cell activation, whereas exenatide enhances glucose-dependent insulin secretion, suppresses glucagon release, slows gastric emptying, and reduces appetite[1][2][4].
