A therapeutic regimen combining dacomitinib, an irreversible pan-human epidermal growth factor receptor (HER) inhibitor, with prophylactic doxycycline to reduce the incidence and severity of skin toxicities associated with dacomitinib treatment. Dacomitinib is indicated as first-line treatment for metastatic non-small cell lung cancer (NSCLC) with EGFR exon 19 deletion or exon 21 L858R substitution mutations. Doxycycline (100 mg twice daily for 4 weeks) has been shown to significantly reduce the incidence of grade ≥2 dermatologic adverse events by 50% and improve quality of life in patients receiving dacomitinib.
