Dactylis glomerata allergen extract, derived from Orchard grass pollen, is a biological product used for the diagnosis and treatment of allergies to this specific grass. For diagnosis, it is administered via skin prick or intradermal tests to elicit a localized IgE-mediated allergic reaction (a wheal-and-flare response) in sensitized individuals. As a therapeutic, it is used in allergen immunotherapy (also known as hyposensitization or allergy shots) to reduce allergic symptoms such as rhinitis and conjunctivitis. This treatment involves administering gradually increasing doses of the extract to induce immune tolerance, which is thought to involve a shift from a Th2-mediated allergic response to a more balanced response with the induction of regulatory T cells and allergen-specific IgG4 blocking antibodies. Clinical studies have also utilized this extract as a controlled challenge agent to investigate the mechanisms of allergic diseases, such as the exacerbation of atopic dermatitis upon environmental exposure.
