Daglutril is an orally active small molecule that acts as a dual inhibitor of neutral endopeptidase (neprilysin) and endothelin-converting enzyme 1. It was developed for the treatment of essential hypertension and congestive heart failure, with additional investigation in patients with type 2 diabetes and nephropathy. By inhibiting both neprilysin and endothelin-converting enzyme, daglutril reduces the production of vasoconstrictor peptides such as endothelin-1 while also preventing the breakdown of natriuretic peptides like atrial natriuretic peptide (ANP), potentially offering antihypertensive effects through multiple mechanisms. Clinical studies have shown that daglutril can lower blood pressure in hypertensive patients with type 2 diabetes and nephropathy but did not significantly affect albuminuria. The drug was originally developed by Solvay Pharmaceuticals and later by AbbVie[2][4][5][6][8].
