Overview

Daidzein 7-O-phosphate (D7P) is a water-soluble phosphate prodrug derivative of the soy isoflavone daidzein. Developed to overcome the poor water solubility and low oral bioavailability of the parent compound, D7P is generated through biotransformation using *Bacillus subtilis* bacteria. The molecule functions as an antagonist of the **cannabinoid receptor 1 (CB1)**, which is notably upregulated in atherosclerotic lesions and endothelial cells subjected to disturbed blood flow. Upon oral administration, D7P is dephosphorylated by alkaline phosphatase to release the active parent compound, daidzein, which exerts vascular protective effects. Preclinical studies have demonstrated that D7P significantly reduces atherosclerotic plaque formation and reverses disturbed flow-induced endothelial dysfunction and endothelial-to-mesenchymal transition (EndMT).
