Overview
Dalfampridine is a small molecule potassium channel blocker used to improve walking in adults with multiple sclerosis (MS). It works by inhibiting voltage-gated potassium channels in demyelinated axons within the central nervous system, thereby enhancing conduction of action potentials and facilitating neuromuscular and synaptic transmission. Dalfampridine is supplied as an extended-release oral tablet and is typically prescribed for patients with MS who have impaired ambulation. The drug was approved by the FDA in 2010 for this indication[1][2][3][7][9].
