Dalotuzumab is a humanized IgG1 monoclonal antibody that targets the insulin-like growth factor 1 receptor (IGF1R). By binding to IGF1R, dalotuzumab prevents the binding of its ligands (IGF1 and IGF2), thereby inhibiting downstream signaling pathways such as PI3K/Akt and Ras/MAPK, which are involved in tumor cell proliferation and survival. This inhibition can result in reduced tumor cell growth and increased apoptosis. Dalotuzumab was developed for the potential treatment of various cancers, including breast cancer, colorectal cancer, multiple myeloma, neuroendocrine tumors, non-small cell lung cancer (NSCLC), pancreatic cancer, and other solid tumors. The drug was originally developed by Pierre Fabre and later licensed to Merck & Co for further development. Clinical trials showed that dalotuzumab could be safely administered alone or in combination with other anticancer agents; however, development has been discontinued for all indications.
