Overview
Dalotuzumab + MK-2206 is a combination therapy investigated in oncology, comprised of **dalotuzumab**, a humanized monoclonal antibody targeting Insulin-like growth factor 1 receptor (IGF-1R), and **MK-2206**, an orally administered small molecule allosteric inhibitor of AKT isoforms (AKT1, AKT2, AKT3). This dual-agent regimen aims to exert vertical pathway inhibition, targeting both upstream (IGF-1R) and downstream (AKT) drivers of cancer cell proliferation, survival, and resistance mechanisms. It has been studied in phase I trials for advanced solid tumors, notably ovarian cancer and colorectal cancer with biomarker-driven populations. The rationale for this combination is that blockade of both IGF-1R and PI3K/AKT/mTOR pathways provides synergistic anti-tumor activity and may help overcome resistance to anti-IGF-1R therapies[1][5].
