Overview
Dalpiciclib is an orally administered, small molecule inhibitor of cyclin-dependent kinases 4 and 6 (CDK4/6), developed primarily for the treatment of breast cancer. By selectively inhibiting CDK4 and CDK6, dalpiciclib blocks phosphorylation of the retinoblastoma (Rb) protein early in the G1 phase of the cell cycle, preventing progression from G1 to S phase and resulting in cell cycle arrest. This mechanism suppresses DNA replication and reduces tumor cell proliferation. Dalpiciclib has been approved in China for advanced or metastatic hormone receptor-positive, HER2-negative breast cancer, and is also being investigated for HER2-positive breast cancer and other solid tumors[1][3][5][7].
