Dalpiciclib + aromatase inhibitor is a combination treatment regimen comprising dalpiciclib—a selective, orally bioavailable cyclin-dependent kinase 4/6 (CDK4/6) inhibitor—and an aromatase inhibitor (most commonly letrozole or anastrozole). This combination is used primarily in the treatment of hormone receptor-positive, HER2-negative advanced or metastatic breast cancer. Dalpiciclib works by inhibiting CDK4 and CDK6, which are crucial for cell cycle progression and tumor cell proliferation. Aromatase inhibitors, such as letrozole and anastrozole, reduce estrogen production, further suppressing hormone-driven tumor growth. Clinical trials (DAWNA-1, DAWNA-2, and others) have shown that this combination significantly improves progression-free survival and objective response rates compared to aromatase inhibitor monotherapy, with a manageable safety profile characterized mainly by hematological toxicities such as neutropenia[1][2][3][4][5][6][8].
