Overview

Dalpicilib (SHR6390) is an orally bioavailable, selective small-molecule inhibitor of cyclin-dependent kinases 4 and 6 (CDK4/6), developed by Jiangsu Hengrui Pharmaceuticals. By inhibiting the CDK4/6-cyclin D complex, dalpicilib prevents the phosphorylation of the retinoblastoma (Rb) protein, leading to G1 phase cell cycle arrest and suppression of tumor cell proliferation. It was first approved in China in 2021 for the treatment of HR-positive, HER2-negative advanced or metastatic breast cancer in combination with endocrine therapy. Ongoing clinical investigations are exploring its efficacy in other solid tumors, including metastatic castration-resistant prostate cancer (mCRPC) in combination with androgen receptor signaling inhibitors like abiraterone acetate.
