Overview

DAN-222 is a novel investigational polymeric nanoparticle drug conjugate that delivers camptothecin, a topoisomerase I inhibitor, directly to tumor tissues. The nanoparticle formulation enhances drug accumulation in tumors via the enhanced permeability and retention (EPR) effect while reducing bone marrow exposure compared to conventional chemotherapy. DAN-222 has demonstrated significant antitumor activity as monotherapy and in combination with PARP inhibitors such as niraparib in preclinical models of breast and ovarian cancer, including both HRD-positive and HRD-negative tumors. Early clinical data indicate that DAN-222 is safe, well tolerated, and shows promising efficacy in patients with metastatic HER2-negative breast cancer. The drug is administered intravenously.
