DAN-311 is a next-generation antibody-drug conjugate (NG-ADC) developed by Dantari for the treatment of HER2-positive and HER2-low breast cancer, built on the company’s Targeted High-capacity Drug Conjugate (T-HDC) platform that enables a substantially higher drug-antibody ratio and tunable payload release compared with conventional ADCs.[1][3][4][7] It targets the HER2 receptor on tumor cells and delivers a topoisomerase I inhibitor payload, aiming to combine potent HER2-directed cytotoxicity with reduced bone marrow exposure and an improved therapeutic index in heterogeneous HER2-expressing tumors.[1][3][6][7] Preclinical data show consistent and durable antitumor activity in HER2-low breast cancer models, and DAN-311 has been advanced from preclinical development toward early clinical evaluation in breast cancer patients who have limited options after current HER2-directed ADCs.[1][3][4][6][9]
