Overview

Danaparoid sodium is a low molecular weight heparinoid anticoagulant composed of a mixture of glycosaminoglycans—primarily heparan sulfate (84%), dermatan sulfate (12%), and chondroitin sulfate (4%)—derived from porcine intestinal mucosa. It acts mainly by binding to antithrombin III, leading to potent inhibition of activated factor X (factor Xa) and, to a lesser extent, activated factor IIa (thrombin), thereby preventing fibrin formation and thrombus development. Danaparoid exhibits minimal cross-reactivity with heparin-induced antibodies, making it suitable for patients with heparin-induced thrombocytopenia (HIT). Its primary indications include the prevention of deep vein thrombosis in surgical patients and off-label use in HIT management. Danaparoid was originally developed by Organon International under the brand name Orgaran but was withdrawn from the US market due to manufacturing shortages; it remains available in some other countries[1][2][3][4][5][7].
