Overview

This drug is a combination of two pharmaceutical agents, danazol and tacrolimus, used together primarily as a second-line treatment for steroid-resistant or relapsed immune thrombocytopenia (ITP). Danazol is a synthetic steroid with antigonadotropic and anti-estrogenic activities that acts by inhibiting pituitary gonadotropin release; it has androgenic properties and is traditionally used in gynecological conditions such as endometriosis. Tacrolimus is an immunosuppressive macrolide that inhibits calcineurin by binding to FKBP-12, thereby suppressing T-lymphocyte activation and IL-2 transcription. The combination leverages the immunosuppressive effects of tacrolimus with the hormonal modulation of danazol to improve outcomes in refractory ITP cases. Tacrolimus blood concentrations can be increased when combined with danazol due to drug interactions affecting metabolism[1][2][3][5][6].
