DANCR201 is a long noncoding RNA (lncRNA) that serves as a protective factor for vascular endothelial function. Research indicates that DANCR201 is significantly downregulated in patients with coronary artery disease, particularly those with vulnerable plaques. Mechanistically, it acts as a molecular sponge for microRNA-216a (miR-216a), thereby inhibiting the miR-216a/NF-κB pathway which is known to promote endothelial dysfunction and inflammation. Overexpression of DANCR201 has been shown to suppress monocyte adhesion to the endothelium, inhibit the expression of pro-inflammatory cytokines (such as IL-1β, ICAM-1, and TNF-α), and enhance endothelial cell proliferation and migration. It is being investigated as a potential therapeutic target for the treatment of atherosclerosis and the prevention of plaque progression and instability.
