Overview

Daniplestim (SC-55494) is a genetically engineered recombinant human interleukin-3 (IL-3) receptor agonist (synthokine) developed to stimulate hematopoiesis. It was designed with multiple amino acid substitutions to bind the IL-3 receptor alpha subunit with significantly higher affinity (approximately 60-fold greater) than native IL-3, thereby enhancing its hematopoietic potency while minimizing dose-limiting inflammatory side effects. Daniplestim was investigated for the mobilization of peripheral blood progenitor cells (CD34+ cells) in patients undergoing high-dose chemotherapy for breast cancer or lymphoma, and for accelerating hematopoietic recovery following myelosuppressive therapy. Development was discontinued in Phase III.
