Overview

Danofloxacin is a synthetic third-generation fluoroquinolone antibiotic used exclusively in veterinary medicine. It exhibits broad-spectrum antibacterial activity against both gram-negative and gram-positive bacteria by inhibiting bacterial DNA gyrase and topoisomerase IV, enzymes essential for DNA replication and cell division. This mechanism results in rapid bactericidal action. Danofloxacin is primarily indicated for the treatment of respiratory diseases in cattle (not intended for dairy cattle producing milk for human consumption), pigs, and chickens. It is approved under brand names such as Advocin (danofloxacin mesylate) for the treatment of bovine respiratory disease (BRD) associated with Mannheimia haemolytica and Pasteurella multocida, as well as other susceptible pathogens[1][2][3][4][5][6]. The drug demonstrates high tissue penetration—especially into lungs—and rapid elimination from the body[2][7]. Developed originally by Pfizer, it is now manufactured by Zoetis among others.
