Overview

Danoprevir + ritonavir is an oral fixed-dose combination of two small molecule antivirals. Danoprevir is a potent inhibitor of the hepatitis C virus (HCV) NS3/4A serine protease, which is essential for viral replication and maturation. Ritonavir acts as a pharmacokinetic booster by inhibiting the metabolism of danoprevir, thereby increasing its plasma concentration and efficacy. The combination was developed primarily for the treatment of chronic hepatitis C infection, particularly in genotype 1 patients, and has demonstrated high sustained virologic response rates when used with other agents such as peginterferon alfa-2a and ribavirin[1][3][4][6]. It was also investigated for COVID-19 pneumonia but development was discontinued for both indications[2]. The drug was originally developed by Roche and later co-developed with Ascletis; it has been marketed in China under the brand name Ganovo since 2018[2][4].
