Dapiglutide is a first-in-class investigational long-acting peptide that acts as a dual agonist of the glucagon-like peptide 1 (GLP‑1) and glucagon-like peptide 2 (GLP‑2) receptors. It is designed for once-weekly subcutaneous administration to treat obesity and address comorbidities associated with low-grade inflammation by improving intestinal barrier function. The drug leverages the weight loss effects of GLP‑1 receptor activation and the gut barrier-enhancing properties of GLP‑2 receptor activation. Clinical studies have shown dapiglutide induces dose-dependent weight loss, improves oral glucose tolerance, delays gastric emptying, reduces plasma glucose and insulin concentrations, promotes intestinal growth, and is generally well-tolerated with a safety profile similar to other incretin-based therapies[1][3][4][5][6][7].
