Dapirolizumab pegol is a novel, investigational, humanized Fc-free polyethylene glycol (PEG)-conjugated antigen-binding (Fab') fragment antibody that targets the CD40 ligand (CD40L), a protein involved in immune system activation. By inhibiting CD40L signaling, dapirolizumab pegol reduces B cell activation and autoantibody production, mitigates type 1 interferon secretion, and attenuates T cell and antigen-presenting cell activation. This broad immunomodulatory effect is designed to address the underlying pathogenesis of systemic lupus erythematosus (SLE). The drug is being developed as an add-on therapy to standard-of-care medications for patients with moderate-to-severe SLE who have persistent or frequently flaring disease activity despite conventional treatment[1][6][8].
