Overview
The combination of daplusiran and tomligisiran is an investigational therapy under development by GSK for the treatment of chronic hepatitis B and hepatitis D infections. This fixed combination consists of two small interfering RNAs (siRNAs), both GalNAc-conjugated, designed to target the hepatitis B virus (HBV) X and S open reading frames (ORFs). The therapy is administered subcutaneously. Clinical studies have shown that this siRNA combination can induce significant reductions in hepatitis B surface antigen (HBsAg) levels in patients with chronic HBV infection. The regimen is being evaluated as part of sequential or combination approaches with other agents such as bepirovirsen to improve rates of functional cure. Daplusiran + tomligisiran is currently in Phase 2 clinical trials for chronic hepatitis B[1][3][6][8].
