Daptomycin is a cyclic lipopeptide antibiotic used to treat serious and life-threatening infections caused by Gram-positive bacteria, including those resistant to other antibiotics such as methicillin-resistant Staphylococcus aureus (MRSA) and vancomycin-resistant enterococci (VRE). It is naturally produced by the soil bacterium Streptomyces roseosporus. Daptomycin acts by inserting into the bacterial cell membrane in a phosphatidylglycerol-dependent manner, causing rapid depolarization of the membrane potential, which leads to inhibition of protein, DNA, and RNA synthesis and ultimately results in bacterial cell death. It is not effective against Gram-negative bacteria or for pulmonary infections due to inactivation by lung surfactant[1][3][5].
