Overview

Daratumumab is a monoclonal antibody that targets CD38, a protein highly expressed on the surface of multiple myeloma and some other malignant cells. By binding to CD38, daratumumab induces cell death through several immune-mediated mechanisms including antibody-dependent cellular cytotoxicity (ADCC), complement-dependent cytotoxicity (CDC), and antibody-dependent cellular phagocytosis (ADCP). It also modulates the immune response by depleting immunosuppressive cells expressing CD38. Daratumumab is primarily indicated for the treatment of multiple myeloma as monotherapy or in combination with other agents, and it is also approved for light chain (AL) amyloidosis in combination regimens. The drug was developed by Janssen and Genmab.
