Overview

This is a four-drug combination regimen used primarily for the treatment of multiple myeloma. It consists of: - **Daratumumab**: a monoclonal antibody that targets CD38 on myeloma cells, inducing cell death through mechanisms such as antibody-dependent cellular cytotoxicity (ADCC), complement-dependent cytotoxicity (CDC), antibody-dependent cellular phagocytosis (ADCP), and direct induction of apoptosis. - **Bortezomib**: a small molecule proteasome inhibitor that disrupts protein degradation in cancer cells, leading to apoptosis. - **Lenalidomide**: an immunomodulatory agent that enhances immune response against tumor cells and has antiangiogenic properties. - **Dexamethasone**: a synthetic glucocorticoid corticosteroid with anti-inflammatory and antitumor effects. The combination is approved for use in newly diagnosed multiple myeloma patients eligible for autologous stem cell transplant as well as those who have received at least one prior therapy. The addition of daratumumab to standard regimens has been shown to significantly improve progression-free survival in clinical trials[1][3][4][8].
