Overview
This combination regimen consists of the anti-CD38 monoclonal antibody daratumumab, the proteasome inhibitor bortezomib, the immunomodulatory agent lenalidomide, the corticosteroid dexamethasone, and the alkylating agent cyclophosphamide. This specific quintuplet list of agents is associated with the MILESTONE trial (NCT05433532) led by the University of Alabama at Birmingham. In the trial, these agents are utilized in two distinct quadruplet induction regimens: DaraVRD (daratumumab, bortezomib, lenalidomide, and dexamethasone) for newly diagnosed multiple myeloma, and DaraVCD (daratumumab, bortezomib, cyclophosphamide, and dexamethasone) for newly diagnosed systemic AL amyloidosis. The study evaluates minimal residual disease (MRD) response-adapted deferral of autologous hematopoietic stem cell transplantation (AHCT) in these patient populations.
