Overview

DCDP (also referred to as DCdP) is a multi-drug combination regimen investigated for the treatment of relapsed and refractory multiple myeloma (RRMM). The regimen consists of four distinct therapeutic agents: daratumumab, a CD38-targeting monoclonal antibody; cyclophosphamide, an alkylating chemotherapy agent; dexamethasone, a corticosteroid; and pomalidomide, an immunomodulatory drug (IMiD). This combination was primarily developed and evaluated by the Canadian Myeloma Research Group (CMRG) in a randomized Phase II clinical trial (NCT02955810). The mechanism of action is synergistic, combining immune-mediated tumor cell lysis via daratumumab, DNA damage through cyclophosphamide, anti-inflammatory and pro-apoptotic signaling from dexamethasone, and the immunomodulatory and anti-angiogenic effects of pomalidomide. Clinical data have demonstrated high overall response rates and significant progression-free survival benefits in heavily pretreated multiple myeloma patients who have failed prior therapies including proteasome inhibitors and lenalidomide.
