Overview
The combination of daratumumab, dexamethasone, ixazomib, and pomalidomide is an investigational quadruplet regimen studied primarily for the treatment of relapsed or refractory multiple myeloma. Daratumumab is a monoclonal antibody targeting CD38 on plasma cells; it induces cell death through immune-mediated mechanisms. Ixazomib is an oral proteasome inhibitor that disrupts protein degradation in cancer cells, leading to apoptosis. Pomalidomide is an immunomodulatory agent that enhances immune response against myeloma cells and inhibits angiogenesis. Dexamethasone is a synthetic glucocorticoid with anti-inflammatory and cytotoxic effects on lymphoid malignancies. This combination aims to leverage complementary mechanisms to improve response rates in patients who have received prior therapies[1][2][4].
