Drug intelligence / Profile preview
Daraxonrasib (Rasonque, RMC-6236) in combination with chemotherapy is a therapeutic regimen under clinical evaluation for the treatment of RAS-mutant cancers, particularly metastatic pancreatic ductal adenocarcinoma (PDAC) and colorectal cancer. Daraxonrasib is a first-in-class, oral, multiselective RAS(ON) tri-complex inhibitor developed by Revolution Medicines. It acts as a molecular glue by first binding to the intracellular chaperone cyclophilin A (CypA) to form a binary complex, which then selectively binds to the active, GTP-bound state of both mutant and wild-type RAS proteins (including KRAS, HRAS, and NRAS). This tri-complex sterically blocks RAS from interacting with downstream effectors like RAF, thereby shutting down the MAPK signaling pathway. Combining daraxonrasib with standard-of-care chemotherapy regimens (such as gemcitabine plus nab-paclitaxel, mFOLFIRINOX, or FOLFOX) is designed to achieve deeper and more durable suppression of RAS signaling and enhance antitumor activity, addressing resistance mechanisms in RAS-addicted gastrointestinal malignancies.
