Overview

Darexaban is an orally active, direct factor Xa inhibitor that was developed by Astellas Pharma for the prevention of venous thromboembolism (VTE) and stroke. It functions by binding directly to the active site of factor Xa, thereby inhibiting the conversion of prothrombin to thrombin and preventing blood clot formation. The drug was investigated in several Phase II and Phase III clinical trials for indications including VTE prevention in patients undergoing major orthopedic or abdominal surgery and stroke prevention in patients with non-valvular atrial fibrillation. However, Astellas announced the discontinuation of darexaban's development in 2011 following a strategic portfolio review.
