M6223 (dargistotug) is an intravenously administered, fully human, Fc-competent monoclonal antibody that acts as an antagonist of T cell immunoreceptor with immunoglobulin and ITIM domains (TIGIT). By inhibiting TIGIT, a co-inhibitory immune checkpoint molecule expressed on T cells and natural killer (NK) cells, M6223 aims to enhance anti-tumor immune responses. The drug is being developed primarily for the treatment of solid tumors and urogenital cancers. It has demonstrated pharmacodynamic effects consistent with TIGIT inhibition in both peripheral blood and tumor tissue. Clinical trials have evaluated its use as monotherapy and in combination with the PD-L1 inhibitor avelumab, particularly in advanced urothelial carcinoma[1][2][4][5][7].
