A fixed-dose or co-administered **combination of daridorexant and famotidine**, consisting of a dual orexin receptor antagonist (DORA) and a histamine H2 receptor antagonist. **Daridorexant** promotes sleep by antagonizing both orexin 1 and orexin 2 receptors, thereby reducing wakefulness and improving sleep architecture, primarily used for the treatment of insomnia. **Famotidine** reduces gastric acid secretion by selectively blocking H2 histamine receptors in the stomach, used primarily to treat conditions like gastroesophageal reflux and peptic ulcer. Coadministration of daridorexant 50 mg and famotidine 40 mg is well tolerated without clinically relevant pharmacokinetic interaction or required dose adjustments for either drug. There is a minor drug interaction classification, but no clinically significant effect has been established; famotidine decreases peak levels of daridorexant (Cmax) without altering its total exposure (AUC)[1][4][5][6][7][8].
