Overview

Darinaparsin is a novel organic arsenical compound consisting of dimethylarsinic acid conjugated to glutathione. Originally developed at The University of Texas MD Anderson Cancer Center, it was designed to provide a more stable and less toxic alternative to inorganic arsenic trioxide. The drug exerts its antineoplastic activity by disrupting mitochondrial function, increasing the production of reactive oxygen species (ROS), and inducing cell cycle arrest and apoptosis in malignant cells. It has demonstrated clinical efficacy particularly in hematologic malignancies. In June 2022, darinaparsin received its first global regulatory approval in Japan for the treatment of patients with relapsed or refractory peripheral T-cell lymphoma (PTCL). The asset was licensed by Alaunos Therapeutics (formerly Ziopharm Oncology) and subsequently out-licensed to Solasia Pharma for development and commercialization in the Pan-Asian region and beyond.
