Darolutamide + niraparib is an investigational combination of two small molecule drugs, each with a distinct mechanism of action, being explored primarily for the treatment of prostate cancer. Darolutamide is a non-steroidal androgen receptor inhibitor that blocks the effect of androgens, thereby slowing the growth and progression of prostate cancer cells. Niraparib is a poly(ADP-ribose) polymerase (PARP) inhibitor that impairs DNA repair in tumor cells, particularly those with homologous recombination repair deficiencies. The rationale for combining these agents lies in targeting both androgen signaling and DNA repair pathways to enhance antitumor efficacy in advanced prostate cancer settings such as metastatic castration-resistant or hormone-sensitive disease[4][8]. Both drugs are orally administered.
