This combination therapy consists of **darunavir** (a protease inhibitor), **ritonavir** (used as a pharmacokinetic booster), and **two nucleoside reverse transcriptase inhibitors (NRTIs)**. It is used as part of highly active antiretroviral therapy (HAART) for the treatment of **HIV-1 infection** in adults and children. **Darunavir** inhibits the HIV-1 protease enzyme, preventing maturation of viral particles. **Ritonavir** strongly inhibits cytochrome P450 3A (CYP3A), increasing plasma levels of darunavir. The NRTIs act by being incorporated into viral DNA by reverse transcriptase, causing chain termination. This triple combination targets HIV replication at two distinct stages: protease-mediated processing and reverse transcription. It is commonly recommended for both treatment-naive and treatment-experienced patients, especially where resistance to other antiretrovirals exists[1][2][3][4][5].
