Overview

This is an investigational combination therapy evaluated in a Phase I trial (NCT01015222) led by the M.D. Anderson Cancer Center. The regimen consists of metronomic (low-dose, frequent) paclitaxel, dasatinib (a BCR-ABL and SRC family tyrosine kinase inhibitor), and bevacizumab (a VEGF-targeting monoclonal antibody), with or without the addition of methylnaltrexone. The study targeted patients with advanced or metastatic malignancies refractory to standard treatments. The rationale behind the combination is to leverage the anti-angiogenic effects of metronomic chemotherapy and bevacizumab alongside the SRC inhibition provided by dasatinib to overcome resistance and inhibit tumor growth and metastasis. Methylnaltrexone was included in specific cohorts to evaluate its potential role in blocking opioid-receptor-mediated signaling that may contribute to tumor progression and angiogenesis.
