Overview
Dasatinib + dacarbazine is a combination of two small molecule anticancer drugs. Dasatinib is a tyrosine kinase inhibitor that targets BCR-ABL, SRC family kinases (SRC, LCK, YES, FYN), c-KIT, EPHA2, and PDGFRβ. It is primarily used to treat Philadelphia chromosome-positive chronic myeloid leukemia (CML) and acute lymphoblastic leukemia (ALL). Dacarbazine is an alkylating agent activated in the liver to form MTIC, which methylates and cross-links DNA, inhibiting DNA, RNA, and protein synthesis. Dacarbazine is mainly used for malignant melanoma and Hodgkin's disease. The combination has been studied in clinical trials for advanced melanoma; dasatinib may sensitize tumor cells to chemotherapy by inhibiting Src kinases[3][4][5][6].
