Dasatinib + imatinib is a combination of two small molecule tyrosine kinase inhibitors used primarily in the treatment of chronic myeloid leukemia (CML), particularly in cases resistant or intolerant to single-agent therapy. Both drugs target the BCR-ABL fusion protein, a constitutively active tyrosine kinase driving CML pathogenesis. Imatinib was the first-in-class inhibitor, while dasatinib is a second-generation agent with greater potency and broader activity against BCR-ABL mutations, including many that confer resistance to imatinib. Dasatinib also inhibits SRC family kinases and other targets such as c-KIT and PDGFRβ. Combination therapy may provide additive or synergistic effects by targeting different leukemic subclones and reducing resistance emergence; it has shown efficacy in patients with TKI-resistant disease without increased toxicity compared to monotherapy[1][2][5][6][8].
