Overview

Dasatinib is an orally available small molecule tyrosine kinase inhibitor used primarily for the treatment of Philadelphia chromosome-positive (Ph+) chronic myeloid leukemia (CML) and acute lymphoblastic leukemia (ALL). It works by inhibiting multiple kinases including BCR-ABL, SRC family kinases, c-KIT, EPHA2, and PDGFRβ. Omeprazole is a proton pump inhibitor that reduces gastric acid secretion by irreversibly inhibiting the H+/K+ ATPase in gastric parietal cells. The combination of dasatinib with omeprazole is generally not recommended because omeprazole significantly reduces the absorption and bioavailability of dasatinib due to increased gastric pH. This interaction can compromise the clinical efficacy of dasatinib in treating cancer[1][4][8].
