**Dasatinib + ponatinib** is a combination of two oral small molecule tyrosine kinase inhibitors (TKIs) used experimentally for the treatment of Philadelphia chromosome-positive (Ph+) leukemias. **Dasatinib** is a multikinase inhibitor that targets BCR-ABL1 and several kinases in the SRC family, while **ponatinib** is a third-generation TKI specifically designed to inhibit BCR-ABL1, including kinase domain mutations such as T315I, which can confer resistance to prior-generation TKIs. The combination offers enhanced inhibition of BCR-ABL1–positive leukemia cells and the potential to suppress the emergence of resistance mutations by targeting overlapping and distinct kinase pathways, including LCK, LYN, FLT3, and CSK[1][3][4][5]. Preclinical data and limited clinical evidence suggest synergistic anti-leukemic effects in vitro and in vivo, leading to more profound inhibition of leukemic cell proliferation and survival compared to either agent alone[1][3].
