DAT-1604 is an orally bioavailable small molecule inhibitor of DNA polymerase theta (Polθ, encoded by the POLQ gene), developed by Danatlas Pharmaceutical. Polθ is a unique DNA polymerase involved in microhomology-mediated end joining (MMEJ), a double-strand break repair pathway that becomes critical in cells with deficiencies in homologous recombination (HR), such as those with BRCA1 or BRCA2 mutations. By inhibiting Polθ, DAT-1604 induces synthetic lethality in HR-deficient tumors. It is currently being evaluated in Phase 1 clinical trials for the treatment of advanced or metastatic solid tumors that are refractory to standard therapies.
