Overview

DAT-2645 is a novel, orally administered small-molecule inhibitor of poly(ADP-ribose) glycohydrolase (PARG), developed by Danatlas Pharmaceuticals (also referenced as Beijing Danqing Pharmaceutical Technology). It is designed to selectively target tumor cells with defects in DNA damage repair (DDR) pathways, particularly those harboring BRCA1/2 loss-of-function mutations. By inhibiting PARG, DAT-2645 exploits synthetic lethality in cancer cells with DDR abnormalities, potentially offering a broad-spectrum therapeutic approach for advanced or metastatic solid tumors. The drug is currently undergoing Phase 1 clinical trials to evaluate its safety, tolerability, pharmacokinetics, pharmacodynamics, and preliminary efficacy in patients with advanced/metastatic solid tumors exhibiting DDR pathway defects[1][2][3][6].
