Datopotamab deruxtecan + capecitabine is a combination of two anti-cancer drugs: datopotamab deruxtecan, an antibody-drug conjugate (ADC) that targets Trop-2 on tumor cells and delivers a DNA topoisomerase I inhibitor payload (DXd), and capecitabine, an orally administered prodrug of 5-fluorouracil (5-FU) that inhibits thymidylate synthase and disrupts DNA synthesis. Datopotamab deruxtecan binds Trop-2 on the cancer cell surface, is internalized, and upon enzymatic cleavage, releases DXd that induces DNA breaks and apoptosis in tumor cells. Capecitabine is converted in the body to 5-FU, which inhibits thymidylate synthase and impairs DNA/RNA synthesis, leading to cell death. This combination is being investigated for potential synergistic effects in advanced solid tumors, especially breast and lung cancers[7].
