Overview
This is a combination regimen of **datopotamab deruxtecan** (Dato-DXd), a TROP2-targeted antibody-drug conjugate (ADC), and **carboplatin**, a platinum-based chemotherapeutic agent. Datopotamab deruxtecan is a humanized monoclonal antibody directed against TROP2 (trophoblast cell surface antigen 2), linked to the topoisomerase I inhibitor exatecan via a cleavable tetrapeptide-based linker. The ADC targets TROP2-expressing tumor cells, is internalized, and, upon cleavage in lysosomes, releases the cytotoxic payload (exatecan) that inhibits DNA replication and induces apoptosis in tumor cells. Carboplatin induces DNA cross-linking, preventing DNA synthesis and promoting cell death. This combination is under investigation for various advanced cancers, particularly non-small cell lung cancer (NSCLC)[2][4][5].
