Drug intelligence / Profile preview
CPX-351 (Vyxeos) is a liposomal encapsulation of cytarabine and daunorubicin in a fixed 5:1 molar ratio. It is designed to optimize the synergistic delivery of these two chemotherapy agents to leukemia cells while reducing systemic toxicity. Cytarabine is a pyrimidine nucleoside analog that inhibits DNA polymerase and DNA synthesis, while daunorubicin is an anthracycline that intercalates DNA and inhibits topoisomerase II. This formulation allows for prolonged exposure and preferential uptake by bone marrow cells. Originally developed by Celator Pharmaceuticals (later acquired by Jazz Pharmaceuticals), CPX-351 is primarily indicated for the treatment of adults with newly-diagnosed therapy-related acute myeloid leukemia (t-AML) or AML with myelodysplasia-related changes (AML-MRC). It is also being investigated for use in pediatric populations with secondary myeloid neoplasms.
