Overview
CPX-351 (Vyxeos) is a liposomal encapsulation of cytarabine and daunorubicin in a fixed 5:1 molar ratio. Developed using the CombiPlex platform, this formulation is designed to maintain the synergistic ratio of the two drugs within the bone marrow for an extended period, enhancing uptake in leukemic cells while reducing systemic toxicity. Cytarabine is a pyrimidine nucleoside analog that inhibits DNA polymerase and is incorporated into DNA, while daunorubicin is an anthracycline that intercalates DNA and inhibits topoisomerase II. It was originally developed by Celator Pharmaceuticals (acquired by Jazz Pharmaceuticals) and is FDA-approved for the treatment of adults and pediatric patients (1 year and older) with newly diagnosed therapy-related acute myeloid leukemia (t-AML) or AML with myelodysplasia-related changes (AML-MRC).
